ABSTRACT

The guidelines for the diagnosis and treatment of hyperthyroidism and other causes of thyrotoxicosis published by the American Thyroid Association (ATA) in 2016,complement and improve the 2011 edition guidelines.The new guidelines put more emphases on the differential diagnosis of thyrotoxicosis;indications of treatment methods including antithyroid drugs (ATD),131I and surgery were revised;a new therapeutic strategy was also put forward for the hyperthyroidism patients being to undergo emergency thyroid surgery.More researches in Asia were included,which provided a certain value for the revision of Chinese guidelines.This article mainly interprets the suggestions for the surgical diagnosis and treatment of hyperthyroidism in 2016 ATA guidelines.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the demonstration ability of 64-multislice spiral computed tomography angiography (MSCTA) with 3-D reconstruction image fusion for the anatomy of perigastric artery.</p><p><b>METHODS</b>From November 2012 to December 2013 in the Nanfang Hospital, a total of 469 patients underwent abdominal 64-MSCTA. 3-D reconstruction technique computed tomography angiography (CTA) was used to reconstruct perigastric arteries. The origin and the course of perigastric arteries were observed. Branching pattern of the hepatic artery was classified by Michels standard, and branching pattern of the right gastric artery was classified into three types according to RGA ramification patterns.</p><p><b>RESULTS</b>Five patients failed to demonstrate hepatic artery because of abnormal development. 3-D reconstruction clearly showed the perigastric arteries in other 464 patients (98.9%). The most common branching pattern of hepatic artery was Michels type I in 346 patients (74.6%). Hepatic artery variation was found in 118 patients (25.4%), including 9 patients of non-Micles type (7.6%). 3-D reconstruction clearly showed the branching pattern of the right gastric artery in 337 patients (72.6%), including 54(16.0%) of proximal pattern, 221 of distal pattern(65.6%), 56 of caudal pattern (16.6%), and 6 of other pattern (3 from splenic artery, 3 from superior mesenteric artery).</p><p><b>CONCLUSION</b>64-MSCTA with 3-D reconstruction can clearly reveal individual perigastric arteries, and can provide guidance for laparoscopic gastrectomy.</p>

ABSTRACT

<p><b>BACKGROUND</b>miR-338-3p is a recently discovered miRNA and is involved in cell differentiation. However, few data are yet available on the aberrant expression of miR-338-3p in human colorectal carcinoma (CRC). This work aimed to investigate the relationship between miR-338-3p expression pattern and clinicopathological features of human CRC and the possible regulative mechanisms.</p><p><b>METHODS</b>The 40 CRC, adjacent nontumorous tissues and 2 human CRC-derived cell lines (SW-480 and SW-620) were collected, respectively, and the total RNA and protein were isolated routinely. The miR-338-3p expression pattern was detected by real-time reverse transcription-polymerase chain reaction (RT-PCR) and Northern blotting. Smoothened (SMO, possible target of miR-338-3p) mRNA and corresponding protein expression pattern were detected by semiquantitative RT-PCR and Western blotting. miR-338-3p expression patterns were compared between nontumor mucosa and CRC samples, graded by progression-related factors. Disease outcome was calculated by Kaplan-Meier survival analysis to determine whether miR-338-3p was related to disease-free survival (DFS) and overall survival (OS) of patients. Moreover, SMO 3'-UTR fragment was PCR amplified from genome DNA of human colon and inserted into a luciferase reporter plasmid. The luciferase reporter plasmid construct was then transfected into CRC cells together with pre-miR-338-3p or anti-miR-338-3p and the luciferase activity in the transfected cells was detected.</p><p><b>RESULTS</b>The expression of miR-338-3p was significantly downregulated in CRCs than those in the adjacent nontumorous tissues, and the value was negatively related to advanced TNM stage and local invasion (P < 0.01). Furthermore, miR-338-3p value was decreased markedly in SW-620 cell line relative to SW-480 (P < 0.01). Low expression of miR-338-3p was associated with unfavorable outcome in DFS but not in OS independent of clinical covariates. Moreover, RT-PCR and Western blotting analysis demonstrated that there was no significant difference in SMO mRNA expression between the corresponding CRCs and nontumorous tissues, whereas SMO protein markedly increased in CRCs (P < 0.01). A significant increase in luciferase activity was detected in CRC cells, which were cotransfected with the luciferase reporter plasmid construct and anti-miR-338-3p (P < 0.01).</p><p><b>CONCLUSIONS</b>miR-338-3p is expressed differentially in CRC and associated with progression and prognosis of CRC. SMO might be a possible target of miR-338-3p, which made it a potential antitumor candidate for treatment and prevention of CRC.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the effect of knocking-down microRNA-221 (miR-221) expression on the radiosensitivity of human colorectal carcinoma cells.</p><p><b>METHODS</b>Human colorectal carcinoma-derived cell line Caco2 was transfected with miR-221 antisense oligonucleotides (anti-miR-221) via Lipofectamine 2000. Real-time quantitative PCR was performed to detect the expression of miR-221 and PTEN mRNA in Caco2 cells. The changes in the protein expression of PTEN in the transfected cells were detected by Western blotting. The cell death after transfection and irradiation was detected by flow cytometry.</p><p><b>RESULTS</b>Transfection with anti-miR-221 caused a significant reduction in miR-221 expression (P<0.05) and up-regulated PTEN protein expression (P<0.05) in Caco2 cells. The percentage of cell death was significantly increased in anti-miR-221 group and anti-miR-221 with irradiation group (P<0.01). Anti-miR-221 significantly enhanced the radiosensitivity of Caco2 cells, which was partially reversed by PTEN-siRNA.</p><p><b>CONCLUSION</b>Anti-miR-221 can enhance the radiosensitivity of colorectal carcinoma cells by up-regulating the expression of PTEN.</p>

ABSTRACT

[Objective] To detect the expression of CD137L mRNA and protein in colorectal cancer tissues and discuss its possible clinical significance.[Methods] To detect the expression of CD137L mRNA in colorectal cancer tissues and adjacent noncancerous tissues by real-time RT-PCR and immunohistochemistry and to analyze the relationship between CD137L expression and clinical features.[Results] The mean level of CD137L mRNA in colorectal cancer tissues is significantly higher than that of adjacent noncancerous tissues (0.035 ± 0.013 vs.0.008 ± 0.005,P < 0.05 ).CD137L mRNA expression in colorectal cancer show no significant differences among different age,sex,tumor location,Dukes classification,histological differentiation,and serum levels of CEA.We selected 8 colorectal cancer tissues and 3 adjacent tissues randomly to analyze the expression of CD137L by immunohistochemistry.CD137L was expressed on tumor cells in colorectal cancer tissues.The expression of CD137L was observed only in colorectal cancer tissues (6/8),but not in adjacent noncancerous tissues (0/3).[Conclusions] The expression of CD137L Mrna in colorectal cancer tissues is higher than that of adjacent noncancerous tissues and CD137 is expressed on tumor cell surface,it might be of biological importance for the progression of colorectal cancer.

Objective To explore the expression and effects of RSp1 and ?-catenin in the intestinal epithelium with intestinal ischemia-reperfusion injury(IIRI)in of mouse.Methods Fifty healthy male kunming mice were randomly divided into control group(n=10)and experimental group(n=40).All mice in control group were only subjected to laparotomy,while the other mice underwent 20 minutes of intestinal mesenteric artery occlusion followed by 6 hours(group A),12 hours(group B),24 hours(group C)and 48 hours(group D)of reperfusion.RT-PCR was used to detect RSpo1 and ?-catenin in small intestine in intestinal ischemia-reperfusion groups and in control group.Results The villous heights of intestinal in experimental groups were significantly lower than that in control group(P
